1、黄连解毒汤对MPTP所致帕金森病小鼠的防治作用作者：段冷昕李瑞芳金瑾王文嘉张杨周秋丽【摘要】目的观察黄连解毒汤（HJD）对1甲基4苯基1,2,3,6四氢吡啶（MPTP）所致C57BL小鼠帕金森病模型的防治作用。方法C57BL小鼠腹腔注射MPTP30mg·kg-1·d-1，连续5d，诱导小鼠帕金森病模型。观察HJD对小鼠肢体运动（爬杆、悬挂、游泳）功能的影响；HPLCECD法检测小鼠脑纹状体内多巴胺（DA）及其代谢产物二羟基苯乙酸（DOPAC）、高香草酸（HVA）的含量；分光光度计法测定小鼠脑组织超氧化物歧化酶（SOD）、谷胱苷肽过氧化物酶(GSHPx)及丙二醛(M

2、DA)的含量。结果与对照组比较，模型组小鼠肢体运动功能明显降低（P<0.05，P<0.01），纹状体内DA、DOPAC及HVA的含量明显减少(P<0.01)，脑组织中SOD及GSHPx酶活力明显降低，但MDA的含量却显著升高(P<0.01)；HJD对小鼠运动功能的减退和脑神经组织的代谢异常有不同程度的调节和改善作用。结论黄连解毒汤对MPTP所致帕金森病小鼠有一定的防治作用。【关键词】黄连解毒汤；帕金森病；MPTP；多巴胺【Abstract】ObjectiveToobservetheprotectiveeffectofHuanglianJiedudeco

4、administrationandsubcutaneousinjection30mg/kgfor5daysattheeighthday.Performanceofanimalsinthepole,tractionandswimmingtestwereobservedat4daysafterthelastinjection.Levelsofdopamine(DA)anditsmetabolites〔3,4dihydroxyphenylaceticacid(DOPAC)andhomovanillicacid(HVA)〕inthestriatumofanimalsweremeasu

5、redbyhighperformanceliquidchromatographywithanelectrochemicaldetector(HPLCECD).Thespectrophotometermethodwasusedtoexaminethelevelsofsuperoxidedismutase(SOD),glutathioneperoxidase(GSHPx)andmalondialdehyde(MDA).ResultsComparingwiththecontrolgroup,themodelgrouphadobviouseffectonthebehavioral

6、performanceoftheanimals,andthedecreasedscoreinthepole,tractionandswimmingtest（P＜0.05orP＜0.01）andthestriatallevelsofDA,DOPACandHVAweresignificantlylower（P＜0.01）,andtheactivityofSODandGSHPxwereimprovedobviouslyinthebrain,butthecontentofMDAwassignificantlyhigher（P＜0.05orP＜0.01）.11Comparingwith
